You can either set calipers at the thickness of a piece that needs measuring and then measure the gap between the points, or you can set the points at a desired dimension and check them against your workpiece as it progresses. Double-sided calipers are good for pre-setting a pair of desired measurements, such as the width and thickness of a bar being drawn out.
